The aircraft, which was carrying 10 passengers and two crew members, crashed during its third attempt at landing at the airport, shortly before 10:00am local time.
It was believed to have caught fire and overturned, with debris scattered across the runway.
A Cork County Council spokesman said three people were dead and two others were trapped in the wreckage.
A statement from the Irish Aviation Authority said "low visibility issues" hampered the plane's attempts to land.
